While the tutorial uses the Function “Filter observations or variables” within Manipulate AnnData, I cannot find it in my instance. Thanks for any suggestions.

Tools evolve over time. This can be complicated! 
To replicate exactly what a tutorial covers, please try to toggle into the same version of a tool that a tutorial is referencing.

For your case, this will be Manipulate AnnData object (Galaxy Version 0.10.9+galaxy0)
