finding mutation, single gene, from RNA-seq data

Hi, I got the review of my paper and they asked me to show that confirmation that one gene did not mutate in the cell lines I used for experiment, as the genetics can change during propagation and database info is insufficient. I was wandering if instead of making new sequencing of this gene I could use sequencing data from my RNA-seq experiment. I have RNA-seq data from those cell lines and I was wanderign if there is any tutorial I could follow for the mutation discovery. All that I found based of variant calling between siblings and parental line which is a different case than mine.
