I am working through the Exome sequencing data analysis for diagnosing a genetic disease tutorial, but with my own set of data. It is WES of a human, so data from only one person. Do I need to create the PED file?

Most of the steps in this tutorial are just a simple baseline variant analysis workflow. The sample comparisons are an extra step layered after that is done.
You can even go as far as to load the data into Gemini. The ped input is optional.

That said, many of the queries in GEMINI concern questions about relationships between samples. With just one individual, those will not be possible. But you can still explore the annotations for the single sample you have!
